室内繁殖松材线虫Bursaphelenchus xylophilus,常规方法是用生长在马铃薯—葡萄糖—琼胶(PDA)平板上的真菌菌丝作为其营养来源,进行单异活体培养。本试验以三角瓶装的玉米粒培养基取代PDA平板,松材线虫的繁殖量提高了20~30多倍。在4℃下,松材线虫在玉米粒上可以存活10个月以上,比在PDA平板上存活时间长。对于仅有少量虫源的松材线虫培养,虫体的表面消毒改在载玻片上的消毒液滴中进行,取代在离心管内消毒的一般作法,可以避免丢失虫体,明显提高培养成功的机率。
Pine wood nematode Bursaphelenchus xylophilus is bred indoors and is routinely cultured in mono-iso cultures using fungal mycelium grown on potato-glucose-agar (PDA) plates as its nutrient source. In this experiment, Triangle bottled corn kernels were used to replace PDA plates, and the reproduction of Bursaphelenchus xylophilus increased by 20 ~ 30 times. At 4 ° C, pine wood nematodes can survive on maize grains for more than 10 months longer than on PDA plates. For the culture of pine wood nematode with only a few insect sources, the surface disinfection of worms was changed to disinfectant droplets on glass slides. Instead of sterilizing in centrifuge tubes, the parasites could be prevented from being lost and the chance of success of culturing was obviously improved .
