目的:研究光照对黄芩悬浮细胞中黄芩苷、黄芩素的积累及相关基因表达的影响。方法:采用HPLC法检测黄芩悬浮细胞中黄芩苷、黄芩素的含量,应用半定量RT-PCR的方法对相关基因的表达进行分析。结果:光照对黄芩悬浮细胞中有效成分的积累有显著的促进作用,与黑暗条件相比,细胞生长13 d后黄芩苷含量显著增长了806.1%,黄芩素含量显著增长了42.5%,PAL基因在光照条件下的转录水平高于在黑暗条件,光照条件下UBGAT同黄芩素显著相关(r=-0.995)。结论:光诱导对黄芩悬浮细胞中有效成分的积累有显著的促进作用,光诱导刺激了与有效成分的积累相关基因(PAL,UBGAT)的表达。
Objective: To study the effects of light on the accumulation of baicalin and baicalein and the expression of related genes in Scutellaria baicalensis suspension cells. Methods: The contents of baicalin and baicalein in Scutellaria baicalensis suspension cells were determined by HPLC. The expression of baicalein and baicalein in Scutellaria baicalensis suspension cells was analyzed by semi-quantitative RT-PCR. Results: Compared with the dark conditions, the content of baicalin increased significantly by 806.1% and the content of baicalein increased significantly by 42.5% after 13 days of cell growth. The expression of PAL gene in the suspension cells of Scutellaria baicalensis was significantly increased UBGAT was significantly associated with baicalein (r = -0.995) under dark conditions and under light. CONCLUSION: Light induction can significantly promote the accumulation of effective components in Scutellaria baicalensis sliver cells, and light-induced stimulation of the expression of genes related to the accumulation of active ingredients (PAL, UBGAT).
