目的研究逍遥散及其各配伍组方对D-半乳糖所致老年性痴呆(Alzheimer′s disease,AD)模型小鼠行为学、形态学及神经递质活性的影响,探讨逍遥散防治AD的作用及其机理,阐释疏肝、养血、健脾相结合的配伍机制。方法 60只SPF级小鼠随机分为空白对照组、模型组、逍遥散全方组、疏肝养血组、疏肝健脾组、健脾养血组6组,每组10只。腹腔注射D-半乳糖复制AD小鼠模型。同时,空白对照组和模型组小鼠每天灌胃生理盐水24mL/kg,逍遥散全方组、疏肝养血组、疏肝健脾组、健脾养血组小鼠分别灌胃各组对应药物水煎液进行对抗治疗,灌胃容积皆为24mL/kg,对应生药量分别为2.485、1.136、1.775及2.059g/kg。第41天进行学习能力训练,第42天进行记忆能力测试。后取脑组织,一半用于测定小鼠脑组织匀浆乙酰胆碱酯酶(acetyl cholinesterase,AchE)、胆碱乙酰转移酶(choline acetyl transefrase,ChAT)及单胺氧化酶(monoamine oxidase,MAO)含量,一半用于病理切片进行形态学观察。结果逍遥散全方组、疏肝养血组、疏肝健脾组、健脾养血组等各治疗组均可提高小鼠学习记忆能力,降低潜伏时间;可提高脑组织匀浆AchE及MAO,降低ChAT;可改善神经元细胞形态、数量及分布,但行为学、形态学及神经递质活性的改善皆以逍遥散全方组最为明显。结论逍遥散能较好的改善AD模型小鼠行为学、形态学及神经酶学,对AD具有较好的防治作用,阐释了疏肝、养血、健脾相结合的配伍方法和组方思路用于防治AD的合理性。
Objective To study the effects of Xiaoyao San and its various components on the behavior, morphology and neurotransmitter activity of D-galactose-induced Alzheimer’s disease (AD) mice and explore the protective effects of Xiaoyaosan on AD Role and mechanism, explain Shugan, nourishing, spleen combination of compatibility mechanism. Methods Sixty SPF mice were randomly divided into 6 groups: blank control group, model group, Xiaoyaosan Decoction group, Shugan Yangxue group, Shuganjianpi group and Jianpi Yangxue group, with 10 in each group. Results Xiaoyao powder group, Shugan Yangxue group, Shuganjianpi group, Jianpi Yangxue group and other treatment groups can improve learning and memory ability, reduce latency; improve brain homogenate AchE and MAO , Reduce ChAT; can improve neuronal cell morphology, quantity and distribution, but behavioral, morphological and neurotransmitter activity were improved Xiaoyao Sanquan group the most obvious. Conclusion Xiaoyao powder can improve the behavior, morphology and neuro-enzymology of AD model mice well and has a good preventive and therapeutic effect on AD, and explain the compatibility method and prescription of combination of Shugan, nourishing and invigorating the spleen For the prevention and treatment of AD rationality.
