目的:体外观察瑞香狼毒醇提物AESC,AESC-1,AESC-2的抗肿瘤作用。方法:采用MTT和SRB法观察AESC,AESC-1,AESC-2对5种肿瘤细胞(A549,NCI-H157,NCI-H460 3种肺癌细胞株,BEL-7402,SK-HEP-1 2种肝癌细胞株)的抑制率,计算IC50,GI50,TGI和LC50。结果:AESC,AESC-1,AESC-2对各细胞株均具有抑制作用,并具有一定的时间依赖性。作用72 h,100,200 mg.L-1各受试药对各细胞株(除A549外)的抑制率在64.82%~92.27%。AESC和AESC-2对NCI-H460和BEL-7402细胞株的IC50<20 mg.L-1,AESC-1对SK-HEP-1细胞株IC50<20 mg.L-1。AESC,AESC-1,AESC-2对敏感细胞株的平均GI50<40 mg.L-1,平均TGI<90 mg.L-1,平均LC50<150 mg.L-1。AESC和AESC-2对敏感细胞株的平均GI50,TGI和LC50值相似。结论:AESC,AESC-1,AESC-2均具有明显抗肿瘤作用,且对不同细胞株敏感性不同;AESC-2与AESC抗肿瘤作用方式相似,AESC的抗肿瘤活性部位可能主要存在于AESC-2。
OBJECTIVE: To observe the antitumor effects of ethanol extracts of Stellera chamaejasme L. in vitro, including AESC, AESC-1 and AESC-2. Methods: MTT and SRB were used to observe the effects of AESC, AESC-1 and AESC-2 on the proliferation of 5 tumor cell lines (A549, NCI-H157 and NCI-H460), BEL-7402 and SK-HEP- Cell lines), IC50, GI50, TGI and LC50 were calculated. Results: AESC, AESC-1 and AESC-2 had inhibitory effect on all the cell lines and had a certain time-dependent manner. The inhibitory rates of various tested drugs (except A549) at 72 h and 100, 200 mg.L-1 ranged from 64.82% to 92.27%. IC50 <20 mg.L-1 for AESC and AESC-2 on NCI-H460 and BEL-7402 cell lines, and IC50 <20 mg.L-1 for AES-1 against SK-HEP-1 cell lines. Average GI50 <40 mg.L-1 for the sensitive cell lines with AESC, AESC-1, and AESC-2, with an average TGI of <90 mg.L-1 and an average LC50 <150 mg.L-1. The average GI50, TGI, and LC50 values for the sensitive cell lines were similar for AESC and AESC-2. Conclusion: AESC, AESC-1 and AESC-2 have obvious anti-tumor effect and different sensitivity to different cell lines. The anti-tumor effect of AESC-2 and AESC are similar, and the anti-tumor activity of AESC may mainly exist in AESC- 2.
