目的检测外周血髓样细胞触发性受体1-(TREM-1)mRNA的表达水平,探讨其在老年全身炎症反应综合征(SIRS)患者中的表达及其意义。方法采用SYBRGreenI实时荧光定量RT-PCR技术,以β-actin为内参照,检测22例老年SIRS患者〔SIRS组,其中多器官功能障碍综合征(MODS组)6例〕和23例健康者(对照组)外周血TREM-1mRNA表达水平。结果 TREM-1和β-actin的熔解曲线均仅有单一峰。SIRS组Ct值(β-actin/TREM-1)为0.930±0.066,正常对照组Ct值为0.846±0.021,2组比较差异具有统计学意义(P<0.01);MODS组Ct值为1.006±0.037,与SIRS组比较差异亦有统计学意义(P<0.05),显著高于正常对照组(P<0.01)。结论应用RT-PCR技术检测TREM-1在外周血中的表达水平简便易行,结果稳定可靠。TREM-1可作为SIRS、MODS早期诊断和病情判断的指标,可能成为SIRS、MODS一个潜在的治疗靶点。
Objective To detect the expression of TREM-1 mRNA in peripheral blood myeloid cells and to explore the expression of TREM-1 mRNA in elderly patients with systemic inflammatory response syndrome (SIRS). Methods Twenty-two elderly patients with SIRS (SIRS group, 6 cases with multiple organ dysfunction syndrome (MODS)) and 23 healthy controls (control group) were detected by real-time fluorescence quantitative RT-PCR with SYBRGreenI and β-actin as internal reference Group) peripheral blood TREM-1 mRNA expression levels. Results The melting curves of TREM-1 and β-actin showed only a single peak. The Ct value was 0.930 ± 0.066 in SIRS group and 0.846 ± 0.021 in normal control group, the difference was statistically significant (P <0.01); the Ct value in MODS group was 1.006 ± 0.037 (P <0.05), which was significantly higher than that of SIRS group (P <0.01). Conclusion The detection of TREM-1 expression in peripheral blood by RT-PCR is simple and easy, and the results are stable and reliable. TREM-1 can be used as an indicator of SIRS, early diagnosis of MODS and judgment of disease condition, and may be a potential therapeutic target for SIRS and MODS.
