目的 以维甲酸诱导急性早幼粒白血病细胞HL60分化为模型 ,用双向电泳技术分析比较维甲酸诱导HL60细胞分化的蛋白表达差异。方法 用细胞生物学的方法分析分化细胞形态上的差异 ,NBT实验检测细胞NBT阳性率。用双向电泳技术分析分化肿瘤细胞蛋白表达差异。结果 分化后细胞形态发生明显变化 ,细胞表面出现突起 ,细胞核变小。分化后细胞NBT阳性率达90 %以上 ,而未分化细胞NBT阳性率低于 5 %。双向电泳技术分析发现有 7个蛋白点只未分化细胞检测到表达 ,而分化细胞未检测到 ;有 14个蛋白点只在分化细胞检测到。结论 双向电泳技术可直接反映细胞蛋白表达变化。
OBJECTIVE: To differentiate HL60 cells induced by retinoic acid into acute promyelocytic leukemia HL60 cells by two-dimensional gel electrophoresis. Methods Cell morphology was used to analyze the morphological differences of differentiated cells. NBT assay was used to detect the positive rate of NBT. Differentiation of Differentiated Tumor Cell Proteins by Two - dimensional Electrophoresis. Results After differentiation, the morphology of the cells changed obviously, the surface of the cells appeared protruding and the nucleus became smaller. After differentiation, the positive rate of NBT was more than 90%, while the positive rate of NBT in undifferentiated cells was less than 5%. Two-dimensional electrophoresis analysis revealed that only seven differentiated cells were detected in undifferentiated cells, whereas differentiated cells were undetectable. Fourteen protein spots were detected only in differentiated cells. Conclusion Two-dimensional electrophoresis can directly reflect the changes of cell protein expression.
