目的 :观察葛根素 (Pur)对大鼠、豚鼠离体主动脉、肺动脉血管平滑肌的作用。方法 :动物急性处死后取出主动脉和肺动脉 ,挂于含 10mlKH液并通入混合气体的浴管中 ,然后观察Pur的作用。结果 :Pur本身对血管无明显作用 ;在去甲肾上腺素 (NA)作用下Pur对大鼠主动脉有促进和抑制收缩的双向作用 ,对肺动脉仅有抑制收缩作用 ,对豚鼠血管具有促进收缩作用 ;Pur可抑制不同浓度KCl引起血管的收缩反应 ,同时不同浓度Pur对KCl的抑制作用存在明显的剂量依赖性 ;Pur与心得安、哌唑嗪有类似作用。结论 :Pur对血管无直接作用 ,但可以明显影响NA、KCl的量效曲线 ,推测Pur具有 β和α受体的阻断作用
Objective: To observe the effects of puerarin on isolated rat aorta and pulmonary artery smooth muscle. METHODS: After the animals were sacrificed after acute death, the aorta and pulmonary artery were removed, suspended in a bath tube containing 10 ml of KH solution and mixed with a gas mixture, and then the effects of Pur were observed. RESULTS: Pur itself had no significant effect on blood vessels; under the action of norepinephrine (NA), Pur had a two-way effect of promoting and inhibiting contraction of rat aorta, only inhibiting contraction of pulmonary artery, and promoting contraction of guinea pig blood vessels. Pur can inhibit the contraction response of blood vessels caused by different concentrations of KCl, and the inhibitory effect of different concentrations of Pur on KCl is dose-dependent; Pur and propranolol and prazosin have similar effects. Conclusion : Pur has no direct effect on blood vessels, but it can significantly affect the dose-response curves of NA and KCl. It is speculated that Pur has the blocking effect of β and α receptors.
