目的采用组织芯片分析PRSS2蛋白在胃癌组织中表达与意义,为获取胃癌诊断相关的蛋白分子提供实验资料。方法首先收集8例胃癌手术切除标本,包括正常胃粘膜、癌旁组织与胃癌组织,选用Westernblotting方法检测PRSS2蛋白在正常胃粘膜、癌旁组织与胃癌组织中的表达;其次运用免疫组织化学染色方法观察组织芯片(包括正常胃粘膜、癌旁和胃癌组织)中PRSS2蛋白在胃癌组织中的表达,分析其临床意义。结果胃癌组织中PRSS2蛋白表达较正常胃粘膜和癌旁组织明显升高,癌旁组织中的表达较正常胃粘膜组织升高;正常胃粘膜、癌旁和胃癌组织中PRSS2蛋白的阳性表达率分别为14.29%(5/34)、26.92%(7/26)和77.78%(63/81),在胃癌组织中高分化、中分化和低分化腺癌的阳性表达率分别为60.00%(3/5)、70.59%(12/17)和81.36%(48/59);PRSS2蛋白的阳性表达与胃癌组织的分化程度也呈现负相关。结论 PRSS2蛋白在胃癌组织中表达较正常胃粘膜和癌旁组织高,癌旁组织中的表达较正常胃粘膜组织高,该蛋白表达与胃癌组织的发生和分化程度有关。
Objective To analyze the expression and significance of PRSS2 protein in gastric cancer by tissue microarray, and provide experimental data for obtaining protein molecules related to gastric cancer diagnosis. Methods Eight specimens of gastric cancer, including normal gastric mucosa, paracancerous tissues and gastric cancer tissues, were collected. Western blotting was used to detect the expression of PRSS2 protein in normal gastric mucosa, paracancerous tissues and gastric cancer tissues. Secondly, immunohistochemical staining To observe the expression of PRSS2 protein in gastric cancer tissues (including normal gastric mucosa, paracancer and gastric cancer tissues), and to analyze its clinical significance. Results The expression of PRSS2 protein in gastric cancer tissues was significantly higher than that in normal gastric mucosa and paracancerous tissues, and the expression of PRSS2 protein in normal gastric mucosa, adjacent tissues and gastric cancer tissues was higher than that in normal gastric mucosa tissues Were 14.29% (5/34), 26.92% (7/26) and 77.78% (63/81), respectively. The positive rates of differentially expressed, moderately differentiated and poorly differentiated adenocarcinoma in gastric cancer were 60.00% (3/5 ), 70.59% (12/17) and 81.36% (48/59), respectively. The positive expression of PRSS2 protein also had a negative correlation with the differentiation of gastric cancer tissues. Conclusion The expression of PRSS2 protein in gastric cancer tissues is higher than that in normal gastric mucosa and para-cancerous tissues. The expression of PRSS2 protein in normal gastric mucosa tissues is higher than that in normal gastric mucosa tissues. The expression of PRSS2 protein is related to the occurrence and differentiation of gastric cancer tissues.
