目的:通过采用HPLC-ELSD法测定不同施肥柴胡中柴胡皂苷a、d的含量,为优选施肥方案提供一定的依据。方法:采用HPLC-ELSD法,色谱柱为Agilent TC-C18(4.6 mm×250 mm,5μm)柱,以水-乙腈(50∶50)为流动相,检测器为蒸发光散射检测器,飘移管温度93℃;空气流速2.5 L.min-1;柱温20℃。结果:确定了柴胡的施肥方案。结论:此方法简单、准确、重复性好,通过柴胡皂苷a、d的变化为优选柴胡施肥方法提供依据。
Objective: To determine the content of saikosaponin a and d in Radix Bupleuri by HPLC-ELSD method, which provides a basis for the optimal fertilization scheme. Methods: HPLC-ELSD method was used. The column was Agilent TC-C18 (4.6 mm×250 mm, 5μm) column with water-acetonitrile (50:50) as mobile phase. The detector was evaporative light scattering detector, drift tube. Temperature 93°C; air flow rate 2.5 L.min-1; column temperature 20°C. Results: The fertilization scheme for Chaihu was determined. Conclusion: This method is simple, accurate and reproducible, and provides the basis for optimizing the fertilization method of Radix Bupleuri by the changes of saikosaponin a and d.
