目的 研究丙型肝炎病毒(HCV)感染外周血单个核细胞(PBMC)的情况及其意义。方法 运用非核素原位杂交法(NISH)和抗生蛋白链菌素-生物素法(SABC)分别检测20例慢性丙型肝炎患者PBMC中的HCV RNA和NS_5抗原。结果 20例患者中有8例(40%)PBMC中HCV RNA呈阳性,其中6例(30%) PBMC中NS_5抗原同时呈阳性。HCV RNA主要分布于胞浆中,而NS_5抗原还可以出现在胞膜上。 HCV RNA和NS_5抗原的阳性细胞比例均很低(分别为≤2%和3%。结论 HCV可以感染PBMC,感染的主要部位为胞浆。 NISH法和PCR法检测PBMC中HCV RNA的结果无显著性差异。
Objective To study the status and significance of infection of peripheral blood mononuclear cells (PBMCs) by Hepatitis C virus (HCV). Methods HCV RNA and NS5 antigen in PBMC from 20 chronic hepatitis C patients were detected by NISH and SABC respectively. Results Eight out of 20 patients (40%) were positive for HCV RNA in PBMCs, of which 6 (30%) were positive for NS5 antigen in PBMCs. HCV RNA is mainly distributed in the cytoplasm, and NS5 antigen can also appear in the cell membrane. HCV RNA and NS_5 antigen were all lower (≤2% and 3%, respectively) .Conclusion HCV can infect PBMC and the main part of infection is cytoplasm.The results of NISH and PCR for detection of HCV RNA in PBMC are not significant Sex differences.
