目的研究脂联素(APN)及其受体(AdipoRs)在正常和1型糖尿病(T1DM)小鼠视网膜中的表达。方法 C57BL/6小鼠随机分为对照组和实验组,通过链脲佐菌素腹腔注射,建立T1DM小鼠模型。建模2月后,分离出对照组和实验组小鼠的视网膜、脉络膜,分别提取总蛋白和总RNA。采用BCA法和ELISA检测视网膜、脉络膜中总蛋白和APN浓度;采用qPCR检测视网膜和脉络膜中APN受体mRNA表达水平;采用Western blotting检测视网膜中AdipoRs的表达情况。结果 APN及其受体在小鼠视网膜、脉络膜中呈现阳性表达;与对照组相比,APN、AdipoRl在实验组小鼠视网膜中表达量上升,AdipoR2表达量无明显变化。结论 APN、AdipoR1可能在Ⅰ型糖尿病视网膜病变病理过程中起重要作用。
Objective To study the expression of adiponectin (APN) and its receptor (AdipoRs) in the retina of normal and type 1 diabetic mice. Methods C57BL / 6 mice were randomly divided into control group and experimental group. The model of T1DM mice was established by intraperitoneal injection of streptozotocin. After 2 months of modeling, the retina and choroid of the mice in the control group and experimental group were separated and the total protein and total RNA were respectively extracted. The retinal and choroidal total proteins and APN concentrations were detected by BCA method and ELISA. The expression of APN receptor mRNA in the retina and choroid was detected by qPCR. The expression of AdipoRs in the retina was detected by Western blotting. Results The expression of APN and its receptor in the retina and choroid of mice showed positive expression. Compared with the control group, the expression of APN and AdipoRl in the retina of experimental mice increased and the expression of AdipoR2 did not change significantly. Conclusion APN and AdipoR1 may play an important role in the pathogenesis of type 1 diabetic retinopathy.
