Notch信号通路通过细胞间受体及配体作用介导信号转导,在系统发育及肿瘤发生、发展等生理病理过程中发挥重要作用。Notch信号通路在脑胶质瘤方面也被广泛研究,Notch配体、受体及其下游基因在脑胶质瘤中都有不同程度的增高,且与胶质瘤级别呈一定的相关性。选择性抑制Notch信号通路,胶质瘤细胞生长受抑、凋亡增加,肿瘤动物模型存活时间延长。最近研究还发现Notch的配体之一DLL4(Deltalike-4)可以抑制肿瘤血管内皮细胞增殖,调控肿瘤血管形成,成为潜在的胶质瘤治疗靶点。
Notch signaling pathway mediates signal transduction through intercellular receptors and ligands and plays an important role in the development of the system and in the pathophysiological and pathological processes such as tumorigenesis and development. Notch signaling pathway is also widely studied in glioma. Notch ligand, receptor and its downstream genes are all increased in gliomas to a certain extent, and have some correlation with glioma grade. Selective inhibition of Notch signaling pathway, inhibition of glioma cell growth, increased apoptosis, survival of tumor animal models prolonged. Recent studies also found that one of the Notch ligands DLL4 (Deltalike-4) can inhibit the proliferation of tumor vascular endothelial cells, regulating tumor angiogenesis, a potential therapeutic target for glioma.
