目的 :探讨外源性重组人钙调素 (recom binant human calmodulin,rh Ca M)对细胞的增殖作用。 方法 :将SP2 /0细胞接种于 2 4孔或 96孔培养板 ,加入不同浓度 rh Ca M、Ca M拮抗剂三氟拉嗪 (TFP)或兔抗 Ca Mγ球蛋白(RACa M)培养 48h后 ,用显微计数法和 MTT比色法检测细胞增殖状况。 结果 :外源性 rh Ca M在一定浓度范围内 (0 .1~ 7.5 μg/ml)与细胞增殖率呈显著正相关 ;Ca M拮抗剂 TFP或兔抗 Ca Mγ球蛋白可抑制细胞增殖。将rh Ca M加入已受 TFP或 RACa M抑制的细胞 ,可恢复正常的细胞增殖功能。 结论 :外源性重组人钙调素可促进细胞增殖。
Objective: To investigate the effect of exogenous recombinant human calmodulin (rhCa M) on the proliferation of cells. METHODS: SP2 / 0 cells were seeded in 24-well or 96-well plates and cultured for 48 h with different concentrations of rhCa M, CaM antagonist trifluoperazine (TFP) or rabbit anti-Ca Mγ globulin (RACa M) The cell proliferation status was detected by micrometer counting and MTT colorimetry. RESULTS: Exogenous rhCa M was positively correlated with cell proliferation rate at a certain range of concentration (0.1-1.5 μg / ml); CaM antagonist TFP or rabbit anti-Ca Mγ globulin could inhibit cell proliferation. Adding rh Ca M to cells that have been inhibited by TFP or RACa M restores normal cell proliferation. Conclusion: Exogenous recombinant human calmodulin can promote cell proliferation.
