目的:寻找白灵侧耳结实相关基因,探寻白灵侧耳结实机理。方法:通过mRNA差异显示技术筛选白灵侧耳菌丝和原基两个发育阶段差异表达的基因片段,经半定量PCR验证后,应用生物信息学方法对片段进行同源性分析并预测其基因功能。结果:共获得8条差异片段,1条与未知蛋白具有40%同源性的基因片段在菌丝中高表达,其余7条在原基中高表达,与细胞色素P450、糖基转移酶、扩展蛋白、糖苷水解酶家族3、核糖体蛋白L29、高半胱氨酸甲基转移酶、未知蛋白的氨基酸同源性为32%~88%不等。结论:差异基因的获得为食用菌结实机理的研究及育种奠定基础。
OBJECTIVE: To search for the genes related to the formation of Pleurotus ostreatus and explore the mechanism of Pleurotus ostreatus. Methods: The differential display gene fragments of Pleurotus citrinopileatus and primordium were screened by mRNA differential display technique. After semi-quantitative PCR validation, the fragments were analyzed by bioinformatics method to predict the gene function . Results: A total of 8 differential fragments were obtained. One gene fragment with 40% homology with the unknown protein was highly expressed in mycelium and the other seven were highly expressed in the primordium, which were highly correlated with cytochrome P450, glycosyltransferase, Glycoside hydrolase family 3, ribosomal protein L29, homocysteine methyltransferase, amino acid homology of unknown protein ranged from 32% to 88%. Conclusion: The acquisition of differential genes laid the foundation for the study of edible fungi ’s mechanism and breeding.
