目的:探索BLCAP基因反义寡核苷酸对人骨肉瘤细胞SOSP-9607的影响,为保肢手术方法作前瞻研究基础。方法:通过细胞培养,测量细胞生长曲线、计算细胞集落形成率、检测细胞周期,观察BLCAP基因反义寡核苷酸对人骨肉瘤细胞系SOSP-9607的影响。结果:BLCAP基因反义寡核苷酸浓度为10μmol/L时,对此细胞的生长速度和集落形成能力有明显促进效应;流式细胞仪检测发现其引起细胞周期变化,对细胞增殖活性有增强作用。结论:首次发现BLCAP基因反义寡核苷酸对人骨肉瘤细胞SOSP-9607有明显的影响,提示BLCAP基因可能作为一种抑癌基因在人骨肉瘤发展过程中起作用。
OBJECTIVE: To explore the effect of BLCAP antisense oligonucleotide on SOSP-9607 human osteosarcoma cells, and to provide a prospective study basis for limb salvage surgery. Methods: The cell growth curve was measured by cell culture, the colony formation rate was calculated and the cell cycle was detected. The effect of BLCAP antisense oligonucleotide on SOSP-9607 cell line was observed. Results: The antisense oligonucleotide of BLCAP gene had a significant promotion effect on the growth rate and colony forming ability of the cells when the concentration of antisense oligonucleotide was 10μmol / L. The flow cytometry showed that it induced cell cycle changes and enhanced the cell proliferation activity effect. CONCLUSION: It was first found that BLCAP antisense oligodeoxynucleotides have a significant effect on human osteosarcoma SOSP-9607, suggesting that BLCAP may play a role as a tumor suppressor gene in the development of human osteosarcoma.
