本实验用~(45)Ca示踪法研究了毛冬青甲素对兔心肌钙离子内外流的影响。结果发现:毛冬青甲素(10~(-5)mol/L)对心肌钙离子内流具有明显促进作用(P<0.05)。这种作用可被钙拮抗剂硝苯吡啶(1.4×10~(-6)mol/L)所抑制(P<0.05),但不被酚妥拉明或普萘洛尔(均10~(-5)mol/L)所阻滞,提示药物的作用可能和α—或β—肾上腺素能受体无关。同时,药物明显增加心肌细胞内的钙离子外流。研究结果提示,毛冬青甲素所具有的正性肌力作用可能是由于药物加快了细胞钙的转运速率所致。
In this study, ~(45)Ca tracer method was used to study the effect of scopolamine on myocardial calcium ion influx in rabbits. The results showed that Maodongqingsu (10~(-5)mol/L) had a significant effect on the influx of calcium in the heart muscle (P<0.05). This effect was inhibited by the calcium antagonist nifedipine (1.4×10 -6 mol/L) (P<0.05), but not by phentolamine or propranolol (10~(-) 5)mol/L) retardation, suggesting that the drug may not be related to α- or β-adrenergic receptors. At the same time, the drug significantly increased the calcium outflow in cardiomyocytes. The results suggest that the positive inotropic effect of Mao Dongqing A may be due to the fact that the drug accelerates the rate of cellular calcium transport.
