目的探讨TCF7L2基因rs290487多态性与辽宁地区2型糖尿病发病的相关性。方法选取2008年12月~2010年7月在中国医科大学附属第四医院内分泌科临床确诊为2型糖尿病的患者255例,提取患者全血基因组DNA,PCR扩增TCF7L2基因单核苷酸rs290487,PCR产物经ACCⅡ酶切后,琼脂糖凝胶电泳分析rs290487的多态性及等位基因rs290487的突变与2型糖尿病发病的相关性。结果 TCF7L2基因rs290487 TT、TC、CC 3个基因型在2型糖尿病组与对照组的分布频率分别为48.24%、46.67%、5.10%和50.74%、40.39%、8.87%,组间差异均无统计学意义(P>0.05);TCF7L2基因rs290487多态位点在两组间的基因型分布符合Hardy-Weinberg平衡定律;Binary Logistic回归校正了性别、年龄、体重指数、血压、血脂等因素后,TCF7L2基因rs290487多态性与2型糖尿病的发生无相关性(P>0.05)。结论辽宁地区人群中TCF7L2基因rs290487多态性与2型糖尿病的发生无相关性。
Objective To investigate the association between rs290487 polymorphism of TCF7L2 gene and type 2 diabetes in Liaoning Province. Methods From December 2008 to July 2010, 255 patients with clinically diagnosed type 2 diabetes in Department of Endocrinology, the Fourth Affiliated Hospital of China Medical University were enrolled. Whole genomic DNA was extracted from patients and PCR was used to amplify TCF7L2 gene single nucleotide rs290487, PCR products were digested by ACC ⅱ, agarose gel electrophoresis analysis rs290487 polymorphism and allele rs290487 mutation and the incidence of type 2 diabetes. Results The distribution frequencies of TT, TC and CC genotypes of rs290487 TCF7L2 gene were 48.24%, 46.67%, 5.10% and 50.74%, 40.39% and 8.87% in the type 2 diabetes mellitus group and the control group respectively, with no statistical difference (P> 0.05). Genotype distribution of rs290487 polymorphism site of TCF7L2 gene was in accordance with Hardy-Weinberg equilibrium law. Binary Logistic regression adjusted for gender, age, body mass index, blood pressure, blood lipids and other factors. TCF7L2 There was no correlation between gene rs290487 polymorphism and type 2 diabetes (P> 0.05). Conclusion There is no correlation between rs290487 polymorphism of TCF7L2 gene and type 2 diabetes in Liaoning population.
