为了探讨ELAC2对乳腺癌细胞MCF-7浸润转移和放射敏感性的影响,利用脂质体转染法将ELAC2基因转入MCF-7细胞,Western blot法检测ELAC2、cyclinB、CDC2和P53蛋白表达水平,细胞划痕法和Boyden小室法检测细胞浸润转移能力,集落形成法检测细胞存活分数,流式细胞仪检测细胞周期。结果表明,成功构建了ELAC2高表达的MCF-7/ELAC2细胞株;ELAC2高表达明显抑制细胞的浸润和转移;细胞受X线照射后,ELAC2高表达使细胞放射敏感性增强,G2期细胞比例增多,同时cyclinB表达降低,P53和Tyr-15磷酸化CDC2表达升高。提示ELAC2高表达可明显抑制细胞浸润和转移;ELAC2高表达使细胞辐射敏感性增加很可能与下调cyclinB、上调P53和Tyr-15磷酸化CDC2蛋白水平有关。
ELAC2 gene was transfected into MCF-7 cells by lipofection method in order to investigate the effect of ELAC2 on invasion and metastasis and breast cancer cell MCF-7 infiltration and radiosensitivity. ELAC2, cyclinB, CDC2 and P53 protein expression levels were detected by Western blot , Cell scratch assay and Boyden chamber method to detect cell invasion and metastasis, colony formation assay to detect cell survival score, flow cytometry cell cycle. The results showed that the ELAC2 highly expressed MCF-7 / ELAC2 cell line was successfully constructed; the high expression of ELAC2 significantly inhibited the infiltration and metastasis of cells; the high expression of ELAC2 enhanced the radiosensitivity of cells after X-ray irradiation, Increased, while cyclinB expression decreased, P53 and Tyr-15 phosphorylated CDC2 expression increased. It suggested that the high expression of ELAC2 could significantly inhibit the cell infiltration and metastasis. The high expression of ELAC2 might increase the radiation sensitivity of the cells, which may be related to the downregulation of cyclinB and the upregulation of phosphorylated CDC2 levels by P53 and Tyr-15.
