目的:优选止痛巴布剂的体外透皮促进剂。方法:采用Franz扩散池,以磷酸缓冲液pH 7.4为接收介质,经白兔腹部离体皮肤渗透,HPLC测定阿魏酸含量,计算药物累积透皮量和累积渗透率,考察透皮吸收促进剂氮酮、N-甲基-2-吡咯烷酮、丙二醇单独使用或混合使用对阿魏酸体外经皮渗透效果的影响。结果:最佳透皮促进剂为3%氮酮+3%丙二醇。N-甲基-2-吡咯烷酮几乎不能增加阿魏酸的透过量,单独加入2%氮酮使阿魏酸透过率增加1.7倍,单独加入3%丙二醇使阿魏酸得透过率增加1.4倍,3%氮酮与3%丙二醇联合使用使阿魏酸的透过率约增加2.2倍。结论:3%氮酮+3%丙二醇作为止痛巴布剂的透皮促进剂效果最好。
Objective: To optimize analgesic cataplasm in vitro transdermal promoting agent. Methods: Ferulic acid was determined by Franz diffusion cell with phosphate buffer pH 7.4 as the receiving medium, exfoliated skin of rabbits in abdomen and the content of ferulic acid by HPLC. The transdermal absorption enhancer Effects of Azone, N-methyl-2-pyrrolidone, and propylene glycol alone or in combination on the transdermal permeation effect of ferulic acid in vitro. Results: The best transdermal enhancer was 3% Azone + 3% propylene glycol. N-methyl-2-pyrrolidone almost did not increase the amount of ferulic acid, ferulic acid alone added 2% 1.7 times increase the transmittance of ferulic acid, 3% propylene glycol alone to increase the permeability of ferulic acid 1.4 Times, 3% Azone and 3% propylene glycol combined use of ferulic acid to increase the transmission rate of about 2.2 times. Conclusion: 3% Azone + 3% propylene glycol is the best transdermal enhancer for analgesic cataplasm.
