目的构建HBsAg转基因小鼠模型,并利用其进行基因治疗研究。方法显微注射外源基因pcDNAHBsAg至FVB小鼠原核,注射胚胎移植到同期发情的假孕受体出生个体,经PCR和Southern检测获得阳性转基因小鼠。通过ELISA方法比较分析转基因小鼠经pcDNAHBsAg质粒基因免疫后抗体产生的情况。结果PCR结合Southern检测到了HBsAg阳性小鼠。基因免疫有抗体产生。结论得到的HBsAg转基因小鼠,基因免疫可诱发转基因小鼠产生抗体。
Objective To construct a mouse model of HBsAg transgenic mice and use it for gene therapy research. Methods Microinjection of exogenous gene pcDNAHBsAg into FVB mice was carried out in prokaryotes. Embryos were transplanted into the pseudopregnant recipients of estrus synchronization. Positive transgenic mice were obtained by PCR and Southern blot. The antibody production of transgenic mice after immunization with pcDNAHBsAg plasmid was compared by ELISA. Results HBsAg positive mice were detected by PCR with Southern blot. Gene immunization with antibody production. Conclusion The HBsAg transgenic mice obtained by gene immunization can induce transgenic mice to produce antibodies.
