目的检测FBLN1在舌鳞状细胞癌(tongue squamous cell carcinoma,TSCC)中表达、启动子区甲基化水平以及甲基化对肿瘤进展过程的影响。方法应用蛋白印记方法检测78例TSCC组织及配对癌旁组织中的表达,应用甲基化特异性聚合酶链反应(Methylation-specic polymerase chain reaction,MSP)技术检测FBLN1基因启动子区甲基化水平。应用SPSS 17.0统计软件进行数据分析,组间比较采用χ2检验和t检验,P<0.05为差异有统计学意义。结果 FBLN1基因在癌组织与正常组织中的相对表达量分别为(0.746±0.326)和(2.353±0.949),差异有统计学意义(t=2.245,P<0.05);FBLN1基因的低表达与肿瘤临床分期及肿瘤的分化程度密切相关(P<0.05)。癌组织基因启动子区甲基化发生率为41.0%(32/78),正常组织甲基化发生率为7.7%(6/78),差异有统计学意义(χ2=19.26,P<0.01)。结论 FBLN1基因的低表达与TSCC患者肿瘤临床分期及肿瘤细胞分化程度密切相关,其低表达与启动子区甲基化有关。
Objective To detect the expression of FBLN1 in tongue squamous cell carcinoma (TSCC), the promoter methylation level and the effect of methylation on tumor progression. Methods 78 cases of TSCC tissues and adjacent tissues were detected by Western blotting. Methylation-specific polymerase chain reaction (MSP) was used to detect the methylation level of FBLN1 promoter . SPSS 17.0 statistical software was used to analyze the data. Chi-square test and t-test were used to compare between groups. P <0.05 was considered statistically significant. Results The relative expression levels of FBLN1 in cancer tissues and normal tissues were (0.746 ± 0.326) and (2.353 ± 0.949), respectively, with significant difference (t = 2.245, P <0.05) Clinical stage and tumor differentiation are closely related (P <0.05). The incidence of methylation in promoter region of cancer tissue was 41.0% (32/78) and that of normal tissue was 7.7% (6/78) (χ2 = 19.26, P <0.01) . Conclusion The low expression of FBLN1 gene is closely related to the clinical stage of tumor and the differentiation degree of tumor cells in TSCC patients. The low expression of FBLN1 gene is associated with the promoter methylation.
