目的:探讨雌激素对大鼠局灶性脑缺血神经细胞凋亡与Bcl-2表达的影响。方法:雌性Wistar大鼠30只均于脑缺血手术前7d切除双侧卵巢,随机等分为假手术组、手术对照组及雌激素用药组。采用大脑中动脉阻断制成局灶性脑缺血模型。缺血2h再灌注24h后断头取脑,应用TUNEL染色及SP免疫组织化学方法观察并测定梗死灶体积、凋亡细胞数及Bcl-2的表达。结果:假手术组未发现脑梗死灶和凋亡细胞及Bcl-2阳性细胞。雌激素用药组较手术对照组脑梗死体积缩小((59.32±8.95)mm3vs(96.93±11.63)mm3,P<0.05),凋亡细胞数减少((96±12)个vs(126±9)个,P<0.05);Bcl-2表达增强(P<0.05)。结论:雌激素通过上调Bcl-2的表达抑制脑细胞凋亡,具有脑保护作用。
Objective: To investigate the effect of estrogen on neuronal apoptosis and Bcl-2 expression after focal cerebral ischemia in rats. Methods: Thirty female Wistar rats were divided into sham operation group, operation control group and estrogen administration group randomly, all at the 7th day before cerebral ischemia surgery. Middle cerebral artery occlusion was used to make focal cerebral ischemia model. The rats were sacrificed at 2 hours after reperfusion for 24 hours after ischemia, and the infarct volume, the number of apoptotic cells and the expression of Bcl-2 were observed by TUNEL staining and SP immunohistochemistry. Results: No cerebral infarction, apoptotic cells and Bcl-2 positive cells were found in sham operation group. Compared with the control group, the volume of apoptotic cells in the estrogen-treated group decreased (59.32 ± 8.95) mm3 vs (96.93 ± 11.63) mm3, P <0.05) , P <0.05), and the expression of Bcl-2 increased (P <0.05). Conclusion: Estrogen can inhibit the apoptosis of brain cells by up-regulating the expression of Bcl-2 and has the protective effect on brain.
