Prabhakaran reported phenoloxidase in M. leprae collected from necropsy spleens, but no reports of O-diphenol oxidase in the serum of leprosy patients have been reported. The authors describe the study of levels of O-diphenol oxidase in serum and cutaneous tissue of leprosy patients. The authors studied 15 healthy individuals, 15 tuberculosis-like leprosy and 15 anemia-type leprosy. O-diphenol oxidase activity was measured using 3,4-dihydroxyphenylalanine (Dopa), tyrosine and catechol as substrates. The reaction mixture contained 1 mL of 0.08 mol Dopa, 2.5 mL 0.01 mol phosphate buffer (pH 6.5) and 0.5 mL impaired skin extract prepared by the Prabhakaran method. The reaction mixture was incubated for 15 minutes at 32 ° C and then read on a Bausch + Lomb Spectronic-20 colorimeter at 480 nm.
