目的:探讨缬草水提物对小鼠腹腔巨噬细胞的白介素-1β(IL-1β)和肿瘤坏死因子-α(TNF-α)mRNA表达的影响。方法:小鼠经缬草水提物灌胃给药1周后,采用逆转录PCR(RT-PCR)方法检测巨噬细胞中的IL-1β和TNF-αmRNA的表达情况。结果:缬草小、中、大剂量组给药后,小鼠腹腔巨噬细胞IL-1βmRNA表达均增高,与空白对照组比较差异显著(P<0.01)。缬草大剂量组给药后,TNF-αmRNA表达增高,与空白对照组比较差异显著(P<0.01)。结论:缬草镇静催眠的机制可能是其增加了小鼠体内IL-1β和TNF-α的含量。
Objective: To investigate the effects of Valeriana officinalis water extract on the expression of interleukin-1β (IL-1β) and tumor necrosis factor-α (TNF-α) mRNA in mouse peritoneal macrophages. METHODS: One week after the mice were intragastrically administered with water extract of Valeriana L., the expression of IL-1β and TNF-α mRNA in macrophages was detected by reverse transcription-PCR (RT-PCR). RESULTS: The IL-1β mRNA expression in peritoneal macrophages increased in the small, medium, and high-dose group, and there was a significant difference compared with the blank control group (P<0.01). In the high-dose group, the expression of TNF-α mRNA was increased, which was significantly different from the blank control group (P<0.01). Conclusion: The mechanism of sedative hypnosis may be that it increases the content of IL-1β and TNF-α in mice.
