目的:研究关附乙酯(DGFA)对大鼠离体心房肌的作用。方法:常规离体器官实验法。结果:DGFA降低了大鼠离体右心房肌的收缩力(P<0.05)并呈浓度依赖性,对右心房的自律性无明显影响;4μmol·L-1DGFA能显著抑制肾上腺素诱发的大鼠离体左房肌的自律性,药前肾上腺素(AD)的浓度为7.92±2.75μmol·L-1,药后10min,AD的浓度升至16.77±6.12μmol·L-1(n=6,P<0.01);4μmol·L-1DGFA在药后10min,使大鼠离体左房的功能性不应期(FRP),由药前的130±12.6ms延长至150±12.6ms(n=6,P<0.05),但对离体大鼠左房肌的兴奋性无明显影响。结论:DGFA的抗心律失常作用可能与其抑制Na+、Ca2+跨膜转运有关。
OBJECTIVE: To study the effects of guanylacetate (DGFA) on rat isolated atrial muscle. Methods: Conventional isolated organ experiments. RESULTS: DGFA reduced the contractility of isolated right atrial muscle in rats (P < 0.05) in a concentration-dependent manner, and had no significant effect on the autoregulation of the right atrium; 4 μmol·L-1DGFA could significantly inhibit the epinephrine-evoked The self-discipline of isolated left atrial muscle in rats, the concentration of preadjuvant adrenaline (AD) was 7.92±2.75 μmol·L-1, and after 10 min, the concentration of AD increased to 16.77±6.12 μmol. L-1 (n=6, P<0.01); 4μmol·L-1DGFA, 10 min after drug administration, left the functional left refractory period (FRP) of rats left atrial, 130±12 before drug. 6ms prolonged to 150±12.6ms (n=6, P<0.05), but there was no significant effect on the excitability of left atrial muscle in isolated rats. Conclusion: The antiarrhythmic effect of DGFA may be related to its inhibition of Na+ and Ca2+ transmembrane transport.
