目的:使用高效液相色谱仪与二极管阵列检测器联用的方法,同时测定柴胡属植物地上部分的十二种黄酮类成分;并用该方法研究黄酮类成分在不同种柴胡中的分布。方法:采用的黄酮对照品分别为儿茶素(1)、5,7,4′-三羟基-芹黄素-6,8-二-碳-β-D-葡萄吡喃糖苷(2)、槲皮素-3-氧-β-D-葡萄吡喃糖基(6→1)-α-L-鼠李吡喃糖苷(3)、山柰素-3,7-二-氧-α-L-鼠李吡喃糖苷(4)、槲皮素-3-氧-α-D-阿拉伯呋喃糖苷(5)、槲皮素-3-氧-α-L-鼠李吡喃糖苷(6)、7-羟基-2,5-二-甲基-色原酮(7)、金合欢素-7-氧-β-D-葡萄吡喃糖基(1→4)-α-L-鼠李吡喃糖苷(8)、山柰素-7-氧-α-L-鼠李吡喃糖苷(9)、槲皮素(10)、山柰素(11)和异鼠李素(12)。色谱分析采用岛津C18色谱柱,检测波长为254 nm,流动相为0.1%的甲酸-水溶液(A)和乙腈(B)梯度洗脱。建立的分析方法还被用于产于不同地区的28个柴胡样品的定量分析。结果:12个黄酮对照品在标准曲线范围内线性关系良好(r2>0.9995),加样回收率在97.10%到102.86%之间,该方法具有良好的精密度、稳定性和重复性。建立的方法被成功用于不同种柴胡地上部分黄酮含量的分析,揭示了柴胡属植物黄酮类成分的分布规律,也为其质量控制提供了依据。
OBJECTIVE: To determine the contents of twelve flavonoids in aerial parts of Bupleurum by using a high-performance liquid chromatography coupled with a diode array detector. The distribution of flavonoids in different species of Bupleurum was studied by this method. Methods: The flavonoid reference substance was catechin (1), 5,7,4’-trihydroxy-apigenin-6,8-di-carbon-β-D-glucopyranoside (2) Quercetin-3-oxy-β-D-glucopyranosyl (6 → 1) -α-L-rhamnopyranoside (3) L-rhamnopyranoside (4), quercetin-3-oxy-aD-arabinofuranoside (5), quercetin- , 7-hydroxy-2,5-dimethyl-chromogen (7), acacetin-7-oxy-β-D-glucopyranosyl Pyranoside (8), behen-7-oxy-α-L-rhamnopyranoside (9), quercetin (10), berylin (11) and isorhamnetin (12). Chromatographic analysis was performed on a Shimadzu C18 column using a gradient elution of formic acid-water (A) and acetonitrile (B) with a detection wavelength of 254 nm and a mobile phase of 0.1%. The established analytical method was also used for quantitative analysis of 28 Bupleurum samples produced in different regions. Results: The calibration curve of 12 flavonoids had a good linearity (r2> 0.9995) within the range of standard curve and the recoveries were between 97.10% and 102.86%. The method has good precision, stability and repeatability. The established method was successfully applied to the analysis of flavonoids in different parts of Bupleurum, revealing the distribution of flavonoids in Bupleurum and providing the basis for its quality control.
