目的:探讨白芍总糖苷(TGP)对类风湿性关节炎(RA)大鼠足爪组织中核转录因子NF-κB/p65蛋白表达的抑制作用和对血清中炎性细胞因子IL-1β、TNF-α、IL-10等水平的影响。方法:建立Ⅱ型胶原诱导的RA大鼠模型。用免疫组化染色法检测足爪组织中NF-κB/p65蛋白的表达。用ELISA法检测RA大鼠血清中IL-1β、TNF-α和IL-10的含量。结果:TGP能以剂量依赖性地下调NF-κB/p65蛋白的表达,降低RA大鼠血清中TNF-α、IL-1β的水平。结论:TGP对RA大鼠炎症的抑制作用,可能与其下调NF-κB/p65蛋白的表达,抑制促炎性细胞因子TNF-α、IL-1β的产生有关。
Objective: To investigate the inhibitory effect of total glucosides of Radix Paeoniae Alba (TGP) on the expression of NF-κB / p65 protein in the paw tissue of rheumatoid arthritis (RA) rats and the effect on the expression of inflammatory cytokines IL-1β, TNF -α, IL-10 levels and other effects. Methods: Type Ⅱ collagen-induced RA rat model was established. Immunohistochemical staining was used to detect the expression of NF-κB / p65 protein in paws. Serum levels of IL-1β, TNF-α and IL-10 in RA rats were detected by ELISA. Results: TGP could down-regulate the expression of NF-κB / p65 protein in a dose-dependent manner and decrease the levels of TNF-α and IL-1β in serum of RA rats. CONCLUSION: The inhibitory effect of TGP on inflammation in RA rats may be related to the downregulation of NF-κB / p65 protein and the inhibition of the production of proinflammatory cytokines TNF-α and IL-1β.
