利用Northerndotblot技术对钙调蛋白(CaM)mRNA转录水平测定观察到:Wistar大鼠在皮下注射AVP(4~8)1h后,酶法所测得的海马组织中游离的CaM蛋白量增至对照组的3.4倍(P<0.05);但同一组织中CaMmRNA转录水平与对照组比较无明显增高,给药3h后,CaMmRNA转录水平达到对照组的2.5倍(P<0.01),同时CaM蛋白量可达对照组的5.8倍(P<0.01),推测这时的CaM可能有两个来源:一是结合性CaM的游离,一是基因表达水平的增高。
The transcription level of calmodulin (CaM) mRNA was detected by using Northerndotblot technique. The amount of free CaM protein in hippocampus measured by enzymatic method was increased to the control group after subcutaneous injection of AVP (4-8) for 1 h in Wistar rats (P <0.05). However, the transcription level of CaM mRNA in the same tissue was not significantly increased compared with that in the control group, and the mRNA level of CaM mRNA was increased 2.5-fold (P <0.01) ). At the same time, the amount of CaM protein reached 5.8 times that of the control group (P <0.01). It is speculated that there are two sources of CaM at this time: one is the dissociation of bound CaM and the other is the increase of gene expression.
