纳米氧化石墨烯对胶质瘤细胞放射增敏的探讨

目的 探讨纳米氧化石墨烯(nGO)颗粒对U87胶质瘤细胞的放射增敏作用及其机制.方法 阿拉玛蓝法检测低浓度nGO联合放射对U87细胞存活的影响.凋亡实验检测U87细胞周期及其凋亡.结果 低浓度nGO对U87细胞生长无明显抑制,nGO联合放射具有抑制细胞生长作用,其抑制效应具有时间依赖性,低浓度nGO对U87细胞即有放射增敏作用.nGO、单纯放射均可诱导细胞凋亡,nGO联合放射组凋亡率显著高于前两组(P<0.01),nGO作用后,细胞周期发生变化,表现G2/M期增加(P<0.05).结论 nGO对人脑胶质瘤细胞株U87具有放射增敏作用,其机制可能为抑制细胞周期,阻滞细胞于G2/M期,并诱导细胞凋亡.
