目的建立尤文肉瘤(Ewing’s sarcoma;ES)血液的差异蛋白质谱,力求从分子水平寻找治疗ES的有效方法。方法通过收集尤文肉瘤患者的全血标本4份(1份/人)和正常健康人群的全血标本4份(1份/人);运用同位素标记相对和绝对定量(Isobaric tags for relative and absolute quantitation,i TRAQ)技术分析尤文肉瘤患者的差异蛋白质。结果共鉴定到了127个差异表达的蛋白,下调的差异蛋白有91个,上调的差异蛋白数是36个。其中热休克蛋白BI(HSPB1)特异表达。结论这是第一次运用全血蛋白表达来鉴定尤文肉瘤的差异蛋白质谱,为尤文的发病机制、疾病进展动态监测及相关治疗奠定了理论基础。
Objective To establish a differential proteomic profile of Ewing’s sarcoma (ES) blood, aiming to find an effective method to treat ES from the molecular level. Methods Four copies of whole blood samples (1 tablet / person) and four healthy volunteers (1 tablet / person) of Ewing’s sarcoma were collected. Isobaric tags for relative and absolute quantitation , i TRAQ) technique to analyze differential proteins in patients with Ewing’s sarcoma. Results A total of 127 differentially expressed proteins were identified, with 91 down-regulated proteins and 36 up-regulated proteins. Among them, heat shock protein BI (HSPB1) is specifically expressed. Conclusion This is the first time that the differential protein profile of Ewing’s sarcoma is identified by the expression of whole blood protein, which lays a theoretical foundation for the pathogenesis of Ewing, the dynamic monitoring of disease progression and related therapies.
