目的:优化生殖营养方多糖的提取条件。方法:设计L_9(3~4)正交实验,采用苯酚-硫酸法显色,以多糖含量为指标,确定碱水提取生殖营养方中多糖的适宜条件。结果:碱水提取多糖的适宜条件:碱水pH=10~11,料液比为1∶70,提取1次,提取时间0.5h,醇沉乙醇浓度75%,该条件下多糖提取率为39.94%。结论:该法多糖提取率高,重现性好,又经济简单,可为生殖营养方多糖提取研究提供参考。
Objective: To optimize the extraction conditions of polysaccharides of reproductive nutrition. Methods: L_9 (3 ~ 4) orthogonal experiment was designed. Phenol - sulfuric acid method was used for the color development. Polysaccharide content was used as an index to determine the suitable conditions for extraction of polysaccharides from reproductive nourishing prescription. Results: The optimal conditions for extracting polysaccharides from alkali water were: alkaline water pH = 10-11, solid-liquid ratio 1:70, extraction time 1, extraction time 0.5h, alcohol-ethanol concentration 75%, polysaccharide extraction rate 39.94 %. Conclusion: The method of polysaccharide extraction rate, reproducibility is good, but also simple and economical, which can provide reference for the extraction of polysaccharides from reproductive nutrition.
