目的探讨解脲脲原体(ureaplasma urealyticum,UU)和微小脲原体(ureaplasma parvum,UP)的生长特性及其分离。方法从妇科门诊随机取266例阴道分泌物标本,通过固、液体培养后应用PCR,基因测序鉴别UU和UP,并筛选出与标准株(ATCC700970)序列相同的UP。取UU标准株T960(CX8=ATCC27618),分别进行培养,绘出生长曲线。结果标本中感染脲原体的有118例,其中UP阳性92例,UU阳性16例,混合阳性10例。固体培养可见大、中、小型的棕色菌落。UU生长周期为22h,UP为30h。液体培养基pH值达到7.5,UU需要13h,UP需要20h。结论临床分离发现UP阳性数高于UU,液体培养中UP的生长周期长于UU,pH值变化速度UP慢于UU,进一步证实了UP和UU是两个不同的种。
Objective To investigate the growth characteristics and isolation of ureaplasma urealyticum (UU) and ureaplasma parvum (UP). Methods A total of 266 vaginal secretions from gynecology clinics were randomly selected. After solid and liquid culture, UU and UP were identified by PCR and gene sequencing, and the same UP sequence as that of the standard strain (ATCC700970) was screened. UU standard strain T960 (CX8 = ATCC27618) was taken and cultured separately, and the growth curve was drawn. Results There were 118 cases of U. urealyticum infection, of which 92 cases were positive for UP, 16 cases were positive for UU and 10 cases were positive for mixture. Solid culture shows large, medium and small brown colonies. UU growth period of 22h, UP for 30h. The pH of the liquid medium reached 7.5, UU required 13h, UP required 20h. Conclusion UP positive number is higher than UU in clinical isolates. The growth period of UP in liquid culture is longer than that of UU, and the change of pH value is slower than that of UU, which further confirms that UP and UU are two different species.
