目的 建立乙氧苯柳胺( E T O F) 软膏皮肤用药后的血药浓度测定方法。方法 采用高效液相联用电化学检测器。结果 E T O F 在625 ~100 μg· L- 1 呈线型关系,日间差5 %~6 % ,日内差21 % ~61 % ,回收率86 % ~98 % ,最低检测限125 pg· L- 1 . E T O F软膏(1 g 含 E T O F150 mg) 分别涂在4 名健康志愿者前臂屈侧4 cm ×4 cm 皮肤后,血液中 E T O F能很快被检出。结论 该方法灵敏度高,重现性好,误差小,能满足 E T O F临床药代动力学研究需要
Objective To establish a method for determination of plasma concentration of topical oestradiol (E T O F) ointment. Methods High performance liquid chromatography with electrochemical detector. Results E T O F showed a linear relationship at 6 25 ~ 100 μg · L -1, with a difference of 5% -6% during the day, an intraday difference of 21% ~ 61% and a recovery of 86% ~ 98% The lowest detection limit of 125 pg · L- 1. E T O F ointment (1 g with E T O F150 mg) was applied to the skin of 4 cm × 4 cm of forearm flexion in four healthy volunteers, and E T O F in the blood can be quickly detected. Conclusion The method has high sensitivity, good reproducibility, small error and can meet the needs of clinical pharmacokinetics of E T O F
