目的:观察安体优I对615小鼠HCa-F肝癌淋巴道转移的影响,探讨其作用机理。方法:生理盐水、干扰素和安体优I分别作用于动物模型,3周后检测转移淋巴结计数及转移率;免疫组化检测VEGF-C和Flt-4,5′-Nase-ALP双重组织化学法检测淋巴管微密度。结果:模型组、α-IFN组和中药组转移淋巴结计数分别为5.88±2.30、4.13±1.64和3.50±1.91个;淋巴结转移率分别为78.6%、61.1%和57.4%;微淋巴管密度分别为25.25±4.59、22.38±4.41和21.75±4.71;VEGF-C的阳性率分别为87.5%、25.0%和25.0%,Flt-4阳性率分别为100.0%、37.5%和50.0%,中药组与模型组比较有显著性差异(P<0.05);结论:安体优Ⅰ具有抑制615小鼠HCa-F肝癌淋巴道转移作用,其机制可能与下调淋巴管生成信号通路VEGF-C及其受体Flt-4的表达,抑制肿瘤淋巴管生成相关。
Objective: To observe the effect of Zadouyou I on lymphatic metastasis of HCa-F hepatocarcinoma in 615 mice and its mechanism of action. Methods: The normal saline, interferon and spironolactone I were used in animal models respectively. After 3 weeks, the numbers of lymph node metastasis and the metastasis rate were detected. The expressions of VEGF-C and Flt-4,5’-Nase-ALP double histochemistry Law detection of lymphatic micro-density. Results: The numbers of metastatic lymph nodes in model group, α-IFN group and TCM group were 5.88 ± 2.30, 4.13 ± 1.64 and 3.50 ± 1.91, respectively. The rates of lymph node metastasis were 78.6%, 61.1% and 57.4% respectively. The lymphatic vessel density were 25.25 ± 4.59,22.38 ± 4.41 and 21.75 ± 4.71 respectively. The positive rate of VEGF-C was 87.5%, 25.0% and 25.0%, respectively. The positive rate of Flt-4 was 100.0%, 37.5% and 50.0% (P0.05) .Conclusion: Antibody-I has inhibitory effect on lymphatic metastasis of HCa-F hepatocarcinoma in 615 mice. The mechanism may be related to the downregulation of VEGF-C and its receptor Flt- 4 expression, inhibition of tumor lymphangiogenesis related.
