超薄切片观察云南烟草丛枝症病害的病叶组织 ,并抽提试验材料的总核酸电泳检测。病组织中检测到与烟草丛枝症病害相关的低分子量RNA ,切片中未观察到病毒粒子和植原体等病原 ,薄壁细胞中发现细胞膜系统的畸变 :质膜体和细胞质泡囊 ,质膜体 (1 0 0 0~ 2 0 0 0nm)结构类似于病毒形成的旁壁体 ,细胞质泡囊(1 0 0~ 2 0 0nm)是含纤维状物的双膜的囊。运用不同盐浓度介质的RNase处理病变细胞组织 ,发现高盐介质的Rnase (2XSSX)不能消解小囊泡中的纤维状物 ,而低盐介质的Rnase (0 0 1XSSC)可以消解小囊泡中的纤维状物。文献显示这些病变是类病毒和某些病毒侵染的特征 ,初步认为这是云南烟草丛枝症病害发病早期细胞内出现小分子核酸侵染的细胞病变特征。
Ultrathin sections were observed in diseased leaf tissues of tobacco witches bush disease in Yunnan Province, and the total nucleic acid electrophoresis of test materials was extracted. In diseased tissues, low molecular weight RNAs associated with tobacco witches’ bush disease were detected. No pathogen such as virions and phytoplasmas was observed in the sections. The aberrations of the cell membrane system were found in the parenchyma cells: plasma membrane and cytoplasmic vesicles, The body (100-200 nm) is similar in structure to the side wall body formed by the virus. The cytoplasmic vesicles (100-200 nm) are fibrous, bicapsuled vesicles. RNase treatment of diseased tissue with different salt medium showed that Rnase (2XSSX) in high-salt medium could not digest fibrils in small vesicles, whereas Rnase (0 0 1XSSC) in low-salt medium could digest small vesicles Fibrous material. The literature shows that these lesions are the characteristics of the virus and certain virus infection, tentatively believe that this is a cell disease characteristic of small molecule nucleic acid infection in the cells early in the onset of tobacco bush disease in Yunnan Province.
