目的:探讨环孢素(CsA)对人脐带间充质干细胞(UC-MSC)增殖、凋亡和细胞周期的影响。方法:培养已被不同浓度CsA处理的UC-MSC,用MTT比色法观察各组UC-MSC增殖,用流式细胞术检测各组细胞凋亡及细胞周期。结果:CsA对UC-MSC增殖无明显抑制作用;CsA可促进UC-MSC凋亡,当CsA药物浓度≥240ng/ml,其细胞凋亡率与药物浓度呈正相关;CsA可改变UC-MSC细胞周期进程,使G0/G1期细胞比例下降,S期细胞比例增高,呈现浓度依赖性。结论:CsA对UC-MSC的细胞学功能无明显负作用。
Objective: To investigate the effects of CsA on the proliferation, apoptosis and cell cycle of human umbilical cord mesenchymal stem cells (UC-MSCs). Methods: UC-MSCs treated with different concentrations of CsA were cultured. The proliferation of UC-MSCs was observed by MTT colorimetric assay. The apoptosis and cell cycle were detected by flow cytometry. CsA can promote the apoptosis of UC-MSCs. When CsA concentration is more than 240ng / ml, the apoptosis rate of CsA is positively correlated with drug concentration; CsA can change the cell cycle of UC-MSCs Process, so that the proportion of G0 / G1 phase cells decreased, the proportion of S phase cells increased in a concentration-dependent manner. Conclusion: CsA has no obvious negative effect on the cytological function of UC-MSC.
