目的 :研究关节软骨细胞在纤维蛋白胶中三维培养时的表现。 方法 :( 1)通过硫酸铵沉淀法从兔血液中提取纤维蛋白原并制备纤维蛋白胶 ;( 2 )在纤维蛋白胶凝成的纤维蛋白中立体培养关节软骨细胞 ,观察其表现并测定生长曲线、倍增时间 ;( 3 )通过透射电镜、AB-PAS染色、 型胶原免疫组化检查等 ,了解细胞生长及软骨基质的分泌情况。结果 :( 1)关节软骨细胞在纤维蛋白中大部分保持卵圆形 ,小部分如贴壁细胞状 ;( 2 )细胞倍增时间在第 1~ 4天为 7.0 d,在第 5~ 7天为 2 .3 d;( 3 )AB-PAS染色 ( +)、 型胶原免疫组化检查 ( +)。结论 :关节软骨细胞能在硫酸铵沉淀法制备的纤维蛋白胶中增殖 ,保持其表和分泌软骨基质
OBJECTIVE: To study the performance of articular chondrocytes in three-dimensional culture of fibrin glue. Methods: (1) Extraction of fibrinogen from rabbit blood by ammonium sulfate precipitation and preparation of fibrin glue. (2) Three-dimensional culture of articular chondrocytes in fibrin-gelatinized fibrin to observe its performance and determine the growth curve , Doubling the time; (3) by transmission electron microscopy, AB-PAS staining, collagen immunohistochemistry, etc., to understand the cell growth and cartilage matrix secretion. Results: (1) The majority of articular chondrocytes remained oval in fibrin, and the small part was like adherent cells. (2) The cell doubling time was 7.0 d on day 1 to 4 and on day 5 to 7 2 .3 d; (3) AB-PAS staining (+), type collagen immunohistochemistry (+). CONCLUSIONS: Articular chondrocytes proliferate in the fibrin glue prepared by ammonium sulfate precipitation to maintain their surface and secrete cartilage matrix
