目的 探讨酪氨酸激酶信息传递途径在成骨细胞增殖及 c- fos表达中的作用。方法 采用阶段性酶消化法分离培养新生大鼠颅盖骨成骨细胞 ,用 MTT法观察成骨细胞增殖 ,用免疫组化 SP法结合计算机图像处理系统检测成骨细胞 c- fos表达。结果 酪氨酸激酶抑制剂 Genistein和 HerbimycinA抑制成骨细胞增殖 ,且两者均部分阻断血清刺激的成骨细胞 c- fos表达。结论 血清刺激和维持成骨细胞增殖及诱导 c- fos表达均涉及酪氨酸激酶信息传递途径
Objective To investigate the role of tyrosine kinase signaling pathway in osteoblast proliferation and c-fos expression. Methods The calvaria osteoblasts were isolated and cultured by phased enzymatic digestion. The proliferation of osteoblasts was observed by MTT assay. The expression of c-fos in osteoblasts was detected by immunohistochemical SP method combined with computer image processing system. Results Tyrosine kinase inhibitors Genistein and Herbimycin A inhibited osteoblast proliferation, and both partially blocked serum-stimulated osteoclast c-fos expression. Conclusion Serum stimulation and maintenance of osteoblast proliferation and induction of c-fos expression involve tyrosine kinase signaling pathway
