目的探讨犬骨髓间充质干细胞(BMMSCs)向血管内皮样细胞分化的机制。方法骨髓穿刺获取犬骨髓,提取单个核细胞,取第2~4代细胞用含VEGF的培养液诱导,对细胞进行形态学观察和相关抗原的免疫荧光鉴定。结果BMMSCs在含VEGF的培养液中诱导,2周后外观呈现鹅卵石样,免疫表型显示vWF阳性表达,透射电镜可见细胞具有血管内皮细胞的特点。结论骨髓间充质干细胞可在体外分化为具有血管内皮细胞特性的细胞。
Objective To investigate the mechanism of differentiation of canine bone marrow mesenchymal stem cells (BMMSCs) into vascular endothelium-like cells. Methods Bone marrow was obtained by bone marrow aspiration, and mononuclear cells were extracted. Cells from passage 2 to passage 4 were induced by culture medium containing VEGF. Morphological observation and immunofluorescence analysis of related antigens were performed. Results BMMSCs were induced in culture medium containing VEGF. After 2 weeks, the appearance of BMMSCs was cobblestone. The immunophenotype showed that the expression of vWF was positive. The cells were characterized by vascular endothelial cells by transmission electron microscopy. Conclusion Bone marrow mesenchymal stem cells can differentiate into cells with the characteristics of vascular endothelial cells in vitro.
