目的观察claudin-6基因在结肠癌、结肠腺瘤及正常结肠黏膜组织中的表达,以探讨其与结肠癌发生发展的关系。方法应用原位杂交法及免疫组织化学法对claudin-6 mRNA及其蛋白在结肠腺瘤(n=20)、结肠癌(n=42)及正常结肠组织(n=10)中的表达进行检测。结果 claudin-6 mRNA及其蛋白在结肠腺瘤及结肠癌组织中的阳性表达率低于结肠正常黏膜,二者在结肠癌组织中的表达又低于结肠腺瘤,组间表达差异有统计学意义(χ~2=9.235,P<0.05;χ~2=8.352,P<0.05);claudin-6 mRNA及其蛋白的表达均与结肠癌组织的分化程度高低、有无淋巴结的转移及浸润肠壁的深度有关(P<0.05),二者在年龄、肿瘤大小及性别组内的表达差异无统计学意义(P>0.05)。结论 claudin-6基因在结肠癌组织中呈低表达,提示claudin-6表达的下调与结肠癌的发生、发展有密切关系。
Objective To investigate the expression of claudin-6 gene in colon cancer, colon adenoma and normal colonic mucosa, and to explore its relationship with the development of colon cancer. Methods The expression of claudin-6 mRNA and its protein in colorectal adenoma (n = 20), colon cancer (n = 42) and normal colon tissues (n = 10) were detected by in situ hybridization and immunohistochemistry . Results The positive expression rate of claudin-6 mRNA and its protein in colorectal adenoma and colon cancer tissues was lower than that in normal colorectal mucosa. The expression of claudin-6 mRNA and protein in colorectal adenocarcinoma was lower than that in colorectal adenoma. There was statistically significant difference between the two groups (Χ ~ 2 = 9.235, P <0.05; χ ~ 2 = 8.352, P <0.05). The expression of claudin-6 mRNA and protein were correlated with the degree of differentiation of colon cancer tissues, lymph node metastasis and infiltration of intestine (P <0.05). There was no significant difference in the age, tumor size and gender between the two groups (P> 0.05). Conclusion The low expression of claudin-6 in colon cancer tissues suggests that the down-regulation of claudin-6 expression is closely related to the occurrence and development of colon cancer.
