目的探讨EphA2/EphrinA1在肾癌中的表达及其与肾癌临床病理特征的关系。方法采用半定量逆转录聚合酶链反应(RT-PCR)和流式细胞术检测22例肾癌组织和12例非肾癌正常肾组织中EphA2、EphrinA1的基因及蛋白表达情况,并对其进行比较和相关分析;利用免疫组织化学法检测68例肾癌组织和24例正常肾组织中EphA2、EphrinA1的表达,分析与肾癌临床病理特征之间的关系。结果肾癌组织中EphA2和EphrinA1的mRAN和蛋白均明显高于正常肾组织(1.60±0.80 vs 0.58±0.19,1.37±0.63 vs 0.91±0.40;424.38±43.14 vs 332.44±42.83,419.44±52.13 vs 325.36±42.33,均P<0.01)。但两种蛋白与其mRAN表达均不存在相关关系(r=0.050,P>0.05;r=-0.171,P>0.05)。在肾癌组织分级不同的各组间,EphA2、EphrinA1蛋白的表达差异有统计学意义(χ2=9.575,P<0.01;χ2=7.947,P<0.05),且随着等级的增加,EphA2、EphrinA1蛋白的表达量也显著增加(r=0.350,P<0.01;r=0.344,P<0.01)。结论EphA2和EphrinA1蛋白高表达于肾癌高分期组、高分级组和淋巴结转移组,其高表达的机制除与基因表达上调有关外,EphA2蛋白降解受阻可能是导致其升高的重要因素。
Objective To investigate the expression of EphA2 / EphrinA1 in renal cell carcinoma and its relationship with the clinicopathological features of renal cell carcinoma. Methods The mRNA and protein expressions of EphA2 and EphrinA1 in 22 cases of renal cell carcinoma and 12 cases of non-renal cell carcinoma were detected by semi-quantitative RT-PCR and flow cytometry. The expression of EphA2 and EphrinA1 in 68 cases of renal cell carcinoma and 24 cases of normal renal tissues were detected by immunohistochemistry. The relationship between the expression of EphA2 and EphrinA1 was analyzed with the clinicopathological features of renal cell carcinoma. Results The mRNA and protein levels of EphA2 and EphrinA1 in renal cell carcinoma were significantly higher than those in normal renal tissues (1.60 ± 0.80 vs 0.58 ± 0.19, 1.37 ± 0.63 vs 0.91 ± 0.40; 424.38 ± 43.14 vs 332.44 ± 42.83, 419.44 ± 52.13 vs 325.36 ± 42.33, all P <0.01). However, there was no correlation between the two proteins and their mRAN expression (r = 0.050, P> 0.05; r = -0.171, P> 0.05). There were significant differences in the expression of EphA2 and EphrinA1 between groups with different grade of renal cell carcinoma (χ2 = 9.575, P <0.01; χ2 = 7.947, P <0.05). With the increase of grade, the expressions of EphA2 and EphrinA1 The protein expression was also significantly increased (r = 0.350, P <0.01; r = 0.344, P <0.01). Conclusions EphA2 and EphrinA1 protein are highly expressed in high-grade, high-grade and lymph node metastasis cases of renal cell carcinoma. The high level of expression of EphA2 and EphrinA1 may be related to the upregulation of gene expression.
