目的:观察清肝化瘀方药阻断大鼠肝癌前病变和癌变的作用及其对信号蛋白ERK的影响。方法:以二乙基亚硝胺长期灌胃诱发大鼠肝癌,以苦参素做对照,进行癌前病变及癌变期HE染色、γ-谷氨酰转肽酶(GGT)组化染色,ERK免疫组化。结果:清肝化瘀方药和苦参素癌前病变及癌变期病理改变较病理组明显减轻。癌前病变期病理组肝组织GGT酶组化染色单位切面病灶数、单位面积内病灶面积、病灶平均面积3项指标显著高于各药物预防组(P<0.05或P<0.01)。单位切面病灶数、单位面积内病灶面积清肝化瘀方药组低于苦参素组(P<0.05)。癌前病变期和癌变期清肝化瘀方药组ERK在肝细胞胞浆和胞核着色较病理组明显减轻,毛细胆管轻微着色,苦参素组ERK在肝细胞异型增生灶、癌巢周围及毛细胆管均着色,但较病理组轻。结论:清肝化瘀方药能阻断二乙基亚硝胺所致大鼠肝癌前病变和癌变的发生,效果优于苦参素。清肝化瘀方药能降低MAPKs通路中通路的活性,抑制细胞过度增殖。
OBJECTIVE: To observe the effect of Qinggan Huayu Recipe on blocking precancerous lesions and carcinogenesis in rats and its effect on signal protein ERK. METHODS: Long-term intragastric administration of diethylnitrosamine induced hepatocellular carcinoma in rats. Matrine was used as a control for precancerous lesions and HE staining and gamma-glutamyl transpeptidase (GGT) histochemical staining for ERK. Immunohistochemistry. RESULTS: Compared with the pathological group, the pathological changes of the precancerous lesions and the cancerous stage of Qinggan Huayu Recipe and oxymatrine were significantly reduced. In the pathological group of precancerous lesions, the GGT enzyme histochemical staining unit number of lesions, the area of lesions per unit area, and the average area of the lesions were significantly higher than the three drug prevention groups (P<0.05 or P<0.01). The number of lesions per unit area and the area of lesions per unit area of Qinggan Huayu formula group were lower than that of oxymatrine group (P<0.05). The colorectal cytoplasm and nucleus of ERK in liver cancer cells were significantly reduced in the precancerous and cancerous stages of the treatment group compared with the pathological group. ERK was slightly stained in the capillary bile ducts, and the ERK in the oxymatrine group and hepatocyte dysplasia lesions and cancer nests were observed. Bile ducts were colored, but lighter than the pathological group. Conclusion: Qinggan Huayu Recipe can block the occurrence of precancerous lesions and carcinogenesis in rats induced by diethylnitrosamine, and the effect is better than that of oxymatrine. Qinggan Huayu Recipe can reduce the activity of pathways in the MAPKs pathway and inhibit excessive cell proliferation.
