目的:观察聚轮烷-喜树碱偶联物对卵巢癌细胞株A2780的抑制作用,以评价其作为抗肿瘤药物的价值。方法:MTT检测偶联物对卵巢癌细胞株A2780的抑制作用;形态学观察经作用后的细胞生长情况;流式细胞仪检测作用后细胞凋亡情况。结果:偶联物对卵巢癌细胞株A2780有显著抑制作用,并呈现量效关系,且其IC50有显著提高;形态学和流式细胞仪检测结果表明,聚轮烷-喜树碱偶联物能够抑制癌细胞生长。结论:聚轮烷-喜树碱偶联物对卵巢癌细胞株A2780有显著的抑制作用,其机制可能是通过诱导细胞凋亡实现的。
Objective: To observe the inhibitory effect of polyrotaxane-camptothecin conjugates on ovarian cancer cell line A2780 in order to evaluate its value as an anti-tumor drug. Methods: The inhibitory effect of the conjugate on the ovarian cancer cell line A2780 was determined by MTT assay. Morphological changes of the cells were observed by flow cytometry. The apoptotic rate was detected by flow cytometry. Results: The conjugate had a significant inhibitory effect on ovarian cancer cell line A2780 with a dose-response relationship and a significant increase in its IC50. Morphological and flow cytometry results showed that the polyrotaxane-camptothecin conjugate Can inhibit the growth of cancer cells. CONCLUSION: Polyrotaxane-camptothecin conjugates have a significant inhibitory effect on ovarian cancer cell line A2780, and its mechanism may be through the induction of apoptosis.
