目的:探讨辛伐他汀联合清肺化痰汤对COPD模型大鼠氧化应激及细胞凋亡的影响作用机制,为慢性阻塞性肺疾病的临床治疗提供新的治疗方案。方法:通过辛伐他汀联合清肺化痰汤对COPD大鼠进行干预,从大鼠的一般状况、肺组织病理变化等多个方面评价其作用;通过测定血清中SOD、GSH、MDA的变化探讨辛伐他汀联合清肺化痰汤对COPD大鼠氧化应激的影响及作用机制;通过测定肺组织凋亡细胞的数目及调亡相关因子(Caspase-3、eNOS、iNOS)蛋白和基因的表达,探讨辛伐他汀对COPD大鼠肺组织细胞凋亡的影响及作用机制。结果:他汀类药物联合清肺化痰汤与COPD患者急性加重期生存率升高有显著关联性,且有利于改善患者预后。结论:COPD患者肺组织细胞凋亡的增加与疾病的发生和进展具有密不可分的关系,值得进行更为深入的研究。
Objective: To investigate the mechanism of simvastatin combined with Qingfei Huatan Decoction on oxidative stress and apoptosis in COPD rats, and to provide a new therapeutic strategy for the clinical treatment of chronic obstructive pulmonary disease. Methods: Simvastatin and Qingfei Huatan decoction were used to intervene COPD rats. The effects of simvastatin combined with Qingfei Huatan Decoction on COPD rats were evaluated. The changes of SOD, GSH and MDA in serum were evaluated from the aspects of general condition and pathological changes of lung tissue. Simvastatin combined with Qingfei Huatan decoction on oxidative stress in COPD rats and the mechanism of action; by measuring the number of apoptotic cells in lung tissue and the expression of protein and gene of apoptosis related factors (Caspase-3, eNOS, iNOS) To investigate the effect of simvastatin on lung apoptosis in COPD rats and its mechanism. Results: The combination of statins and Qingfei Huatan Decoction significantly correlated with the survival rate of COPD patients at acute exacerbation stage, and was beneficial to improve the prognosis of patients. Conclusion: The increased apoptosis of lung tissue in patients with COPD is closely related to the occurrence and progression of the disease, which deserves further study.
