目的优化水相反应体系中的共轭亚油酸与薄荷醇的酶催化合成反应条件。方法根据中心组合试验设计原理,采用五因素五水平的响应曲面分析法,以反应转化率(%)为响应值作响应面和等高线图,考察了温度、反应时间、酶量、底物物质的量比、水量的影响。结果优化后的酶催化反应条件为:温度35℃、反应时间36 h、酶量40%、底物物质的量比为1,水量30%。结论本文所建立的数学模型显著(P<0.05),可用来对水相体系中的共轭亚油酸-薄荷醇酯的酶催化合成反应进行分析和预测。
Objective To optimize the conditions for the enzymatic synthesis of conjugated linoleic acid and menthol in aqueous reaction system. Methods According to the design principle of central composite experiment, response surface and contour maps were obtained by response surface conversion (%) as the response value using five factors and five levels of response surface methodology. Results The optimal reaction conditions were: reaction temperature 35 ℃, reaction time 36 h, enzyme 40%, substrate ratio 1, water 30%. Conclusion The mathematical model established in this paper is significant (P <0.05), which can be used to analyze and predict the enzyme catalyzed synthesis of conjugated linoleic acid-menthol ester in aqueous system.
