目的探讨β2肾上腺素受体(β2-adrenoceptor,β2AR)基因(β2AR)Arg16Gly多态性对健康青年血脂及载脂蛋白比值的影响及在高糖低脂(high-carbohydrate/low-fat,HC/LF)膳食诱导的血脂及载脂蛋白比值变化中的作用。方法56例健康志愿者,给予7d平衡膳食和6dHC/LF膳食,于第1、8天及第14天取12h空腹静脉血,测定血脂和载脂蛋白(apolipoprotein,apo)AI、B100水平,计算TG/HDL-C、log(TG/HDL-C)、TC/HDL-C、LDL-C/HDL-C、apoAI/apoB100值。提取基因组DNA,聚合酶链反应-限制性酶切法分析Arg16Gly多态性。结果不同性别AA基因型受试者与G等位基因携带者之间血脂比值的基础值差异无统计学意义(P>0.05)。平衡膳食后及HC/LF膳食后,无论是受试人群整体还是将男女分组分析,AA基因型受试者与G等位基因携带者之间血脂及载脂蛋白比值亦无明显差异(P>0.05)。与HC/LF膳食前相比,HC/LF膳食后,在受试人群整体,AA基因型受试者TG/HDL-C(P=0.017)、log(TG/HDL-C)(P=0.031)、apoAI/apoB100(P=0.006)显著升高,LDL-C/HDL-C和TC/HDL-C显著降低(P<0.01),而G等位基因携带者仅LDL-C/HDL-C和TC/HDL-C显著降低(P<0.01)。男女分组分析发现,与HC/LF膳食前相比,男性受试组中HC/LF膳食后LDL-C/HDL-C(AA基因型:P=0.035,G等位基因:P=0.002)和TC/HDL-C(AA基因型:P<0.01,G等位基因:P<0.01)在AA基因型受试者与G等位基因携带者中均显著下降,apoAI/apoB100仅在AA基因型显著升高(P=0.003);女性受试组中,HC/LF膳食后LDL-C/HDL-C(AA基因型:P=0.011,G等位基因:P=0.043)和TC/HDL-C(AA基因型:P<0.01,G等位基因:P<0.01)在AA基因型受试者与G等位基因携带者均显著下降,TG/HDL-C(P=0.001)和log(TG/HDL-C)(P=0.001)仅在AA基因型较膳食前显著升高。结论β2肾上腺素受体基因Arg16Gly变异能抑制HC/LF膳食诱导的健康青年女性TG/HDL-C和log(TG/HDL-C)升高,而HC/LF膳食诱导的TG/HDL-C和log(TG/HDL-C)改变在该位点野生型(AA基因型)和变异携带者(G等位基因)健康青年男性之间没有明显差异,可能与apoAI/apoB100有关。
Objective To investigate the effect of Arg16Gly polymorphism of β2-adrenoceptor (β2AR) gene on serum lipids and apolipoproteins in healthy volunteers and the effect of high-carbohydrate / low-fat (HC / LF) diet-induced changes in blood lipids and apolipoprotein ratio role. Methods Fifty-six healthy volunteers were given 7d balanced diet and 6dHC / LF diet. On the 1st, the 8th and the 14th days, the fasting venous blood samples were collected and the levels of AI and B100 in serum lipids and apolipoprotein (apo) were measured. TG / HDL-C, log (TG / HDL-C), TC / HDL-C, LDL-C / HDL-C, apoAI / apoB100 values. The genomic DNA was extracted and the Arg16Gly polymorphism was analyzed by polymerase chain reaction - restriction enzyme digestion. Results There was no significant difference in the basic lipid ratio between AA genotypes and G allele carriers (P> 0.05). After balanced diet and HC / LF diet, there was no significant difference in serum lipid and apolipoprotein ratio between subjects with AA genotype and those with G allele, both in the overall study population and in the subgroups of men and women (P> 0.05). After HC / LF diet, the levels of TG / HDL-C (P = 0.017), log (TG / HDL-C) ), ApoAI / apoB100 (P = 0.006), LDL-C / HDL-C and TC / HDL-C were significantly lower (P <0.01) And TC / HDL-C significantly decreased (P <0.01).
