目的探讨hsa-miR-26b表达沉默对脂肪细胞蛋白分泌的影响。方法将hsa-miR-26b表达沉默组及对照组慢病毒感染人脂肪前体细胞,在诱导分化过程中收集不同时间点的细胞培养基上清液,采用ELISA法检测IL-6和瘦素(leptin)的分泌量。结果在细胞分化过程中,hsa-miR-26b表达沉默组IL-6与leptin表达量均较对照组降低(P<0.01或P<0.05)。结论降低hsa-miR-26b表达在一定程度上可以改善人脂肪细胞炎症状态及胰岛素抵抗,为肥胖治疗提供潜在靶点。
Objective To investigate the effect of hsa-miR-26b silencing on the secretion of adipocyte protein. Methods The hsa-miR-26b-silenced and control lentivirus-infected human preadipocytes were used to collect supernatants of cell culture medium at different time points during differentiation. IL-6 and leptin leptin secretion. Results The expression of IL-6 and leptin in the hsa-miR-26b silencing group was lower than that in the control group (P <0.01 or P <0.05) during cell differentiation. Conclusions Reducing the expression of hsa-miR-26b can improve the inflammatory status of human adipocytes and insulin resistance to a certain extent, and provide a potential target for the treatment of obesity.
