目的 探讨急性淋巴细胞白血病 (ALL)和慢性髓性白血病 (CML)易感性与HLA DRB1基因多态性之间的关联性 ,找出急性淋巴细胞白血病和慢性髓性白血病的易感基因。方法 采用序列特异性引物聚合酶链反应 (PCR SSP)DNA分型技术对 5 6例ALL患者、48例CML患者和 10 5例健康对照进行了HLA DRB1基因分型。结果 ALL患者组与HLA DR7基因关联 ,基因频率为 2 4.1% ,RR=2 .5 6 ,χ2 =7.34 ,P <0 .0 1;CML患者组与HLA DR4基因关联 ,基因频率为 2 2 .9% ,RR =5 .0 76 ,χ2 =17.88,P <0 .0 1;其他等位基因频率在实验组与对照组间差异无显著性。结论 提示在河南汉族人群中 ,HLA DR7与ALL有关联 ,HLA DR4与CML有关联。
Objective To investigate the association between susceptibility to acute lymphoblastic leukemia (ALL) and chronic myeloid leukemia (CML) and HLA DRB1 gene polymorphisms and to identify susceptibility genes for acute lymphoblastic leukemia and chronic myeloid leukemia. METHODS: HLA DRB1 genotype was performed on 56 patients with ALL, 48 patients with CML, and 105 healthy controls using sequence-specific primer polymerase chain reaction (PCR SSP) DNA typing technique. Results The ALL patient group was associated with HLA DR7 gene, the gene frequency was 2 4.1%, RR = 2.56, χ2 = 7.34, P <0.01; CML patient group was associated with HLA DR4 gene, gene frequency was 2 2. 9%, RR = 5.06, χ2 = 17.88, P <0.01; other allele frequencies were not significantly different between the experimental group and the control group. Conclusion It is suggested that in Henan Han population, HLA DR7 is associated with ALL, and HLA DR4 is associated with CML.
