目的探讨集落刺激因子1受体(CSF-1R)对人鼻咽癌6-10B细胞细胞周期素(cyclin)D1表达及其增殖的影响。方法选取人鼻咽癌6-10B细胞,体外利用慢病毒构建的CSF-1R过表达载体LV-CSF1R(16957-1)转染到人鼻咽癌6-10B细胞中(转染组),未转染CSF-1R慢病毒载体的鼻咽癌6-10B细胞为对照组。采用RT-q PCR法及Western blotting法检测转染组及对照组细胞CSF-1R、cyclin D1的表达情况,CCK-8法检测各组细胞活力的改变情况。结果转染组细胞CSF-1R mRNA及其蛋白表达水平明显高于对照组(P均<0.05)。转染组鼻咽癌6-10B细胞活力较对照组明显增高(P<0.05)。结论过表达CSF-1R可以通过调节细胞周期蛋白cyclin D1来促进鼻咽癌6-10B细胞的恶性生长。
Objective To investigate the effects of CSF-1R on the expression of cyclin D1 and its proliferation in human nasopharyngeal carcinoma 6-10B cells. Methods Human nasopharyngeal carcinoma 6-10B cells were selected and transfected into 6-10B human nasopharyngeal carcinoma cells (transfection group) with lentivirus vector LV-CSF1R (16957-1) Nasopharyngeal carcinoma 6-10B cells transfected with CSF-1R lentiviral vector served as control group. RT-q PCR and Western blotting were used to detect the expression of CSF-1R and cyclin D1 in transfected and control cells. The changes of cell viability were detected by CCK-8 assay. Results The expression of CSF-1R mRNA and protein in transfected cells was significantly higher than that in control group (all P <0.05). Transfection of nasopharyngeal carcinoma cell line 6-10B was significantly higher than the control group (P <0.05). Conclusion Overexpression of CSF-1R can promote the malignant growth of nasopharyngeal carcinoma 6-10B cells by regulating the expression of cyclin D1.
