目的:研究缺氧诱导因子-1α(HIF-1α)与抑癌基因p53(p53)突变型在甲状腺乳头状癌(PTC)中的表达情况,探讨HIF-1α和p53在PTC组织中的意义及其与临床病理参数的关系。方法:选取2016年3月至2018年3月盘锦辽油宝石花医院外科手术切除并存档于病理科的原发性PTC组织60例及癌旁正常甲状腺组织20例,采用免疫组织化学法及实时定量基因扩增荧光检测法(qPCR)检测两组HIF-1α与p53蛋白及mRNA水平,分析二者相关性及其与临床参数之间的相关性。结果:PTC组织中HIF-1α和p53蛋白阳性表达均高于癌旁正常甲状腺组织[66.7%(40/60)比6/20、70.0%(42/60)比6/20](n P<0.05),且HIF-1α和p53表达呈正相关(n r=0.849,n P=0.001)。PTC组织中HIF-1α和p53 mRNA水平高于癌旁正常甲状腺组织(n P<0.05),且二者表达呈正相关(n r=0.594,n P=0.002)。PTC组织中HIF-1α和p53蛋白表达强度分别与临床分期(n r=0.386,n P=0.003;n r=0.463,n P0.05)。n 结论:HIF-1α和p53在PTC中表达上调,与临床分期和淋巴结转移呈正相关,二者可能在PTC的发生、发展和转移中协同发挥促进作用。“,”Objective:To investigate the expression of the hypoxia-inducible factor-1α(HIF-1α) and tumor suppressor gene p53 (p53) in papillary thyroid carcinoma (PTC), and to explore the significance of HIF-1α and p53 and its relationship with clinicopathological parameters in PTC.Methods:Immunohistochemistry and fluorescence quantitative real-time quantitative (qPCR) were used to detect the levels of protein and mRNA expression of HIF-1α and p53 in PTC and adjacent normal tissues, and explore the correlation and clinical parameters analysis between them.Results:The expression rates of HIF-1α and p53 protein in PTC were higher than those in adjacent normal tissues[66.7%(40/60) vs. 30.0%(6/20),70.0%(42/60)vs.30.0%(6/20)] (n P<0.05), and there was positively correlated (n r=0.849, n P=0.001). The levels of HIF-1α and p53 mRNA in PTC were higher than those in adjacent normal tissues (n P<0.05), and there was positive correlation (n r=0.594, n P=0.002). The expression of HIF-1α and p53 protein in PTC were positively correlated with clinical stage (n r=0.386, n P=0.003; n r=0.463 n P0.05).n Conclusions:The expression of HIF-1α and p53 is up-regulated in PTC, which is positively correlated with clinical stage and lymph node metastasis. Both of them may play a synergistic role in the occurrence, development and metastasis of PTC.
